Effect of Steroid and Gonadotropin on the Cumulus Expansion and Oocyte Maturation in Vitro

Abstract
In order to study hormonal stimulation of cumulus expansion and oocyte maturation, oocyte cumulus complexes isolated from graffian follicle of ICR mice for 48 hrs after PMSG in injection.
Oocyte-cumulus complexes were cultured in the presence of various hormones for 17hrs at 37¡ÉCO2 incubators.
@ES The results obtained in these experiments were summarized as follow;
@EN 1. Cumulus expansion was stimulated in vitro FSH, Humegon hCG (Chorulon), LH in M16 medium containing FCS.
2. The maximal stimulating concentration of FSH was found to be about 0.01§¶/ml.
3. Neither Estradiol and hCG(Sigma, U.S.A.) stimulated cumulus expansion.
4. The stimulation of cumulus expansion by FSH, hCG(Choluon)or Humegon required the presence of FCS.
5. Oocyte maturation did not appear to be affected by progesterone.
6. The majority of the follicular oocytes with compacted cumulus cells existed in GV stage while those with dispersed cumulus cell or without cumulus cell existed between GVBD and MII stage.
